چکیده

objective(s) the main objective of this study was to investigate the status of chromosome stability in 3 human-mouse hybridoma cell lines over a period of time in various passages. materials and methods metaphase spreads from 3 human-mouse cell lines (hf2x653, spmo-4 and f3b6) that had been cultured in 4 successive passages, from 1 to 4 weeks, were prepared and analyzed. metaphase chromosomes stained in giemsa and a fluorescent dye, hoechst 33258, for differential staining. this staining was performed for differentiating human and mouse chromosomes. results numerical chromosome analysis showed that although in successive passages the total number of chromosomes in hybridoma cells remained unchanged, some changes occurred in the number of human and mouse metacentric and acrocentric chromosomes during different passages. these changes were detectable, using fluorescence staining method. conclusion since one of the main uses of human-mouse hybridoma cells is producing monoclonal antibody, chromosomal instability in these cells causes the loss of human chromosomes coding the antibody of interest occasionally. therefore, cytogenetical analysis and characterization of these cells, especially by using the appropriate ways of chromosomal identification, is essential prior to use.

Passaging HuES human embryonic stem cell-lines with trypsin.

In this video we demonstrate how our lab routinely passages HuES human embryonic stem cell lines with trypsin. Human embryonic stem cells are artifacts of cell culture, and tend to acquire karyotypic abnormalities with high population doublings. Proper passaging is essential for maintaining a healthy, undifferentiated, karyotypically normal HuES human embryonic stem cell culture. First, an expa...

Chromosomal variation in lymphoblastoid cell lines.

Tens of thousands of lymphoblastoid cell lines (LCLs) have been established by the research community, providing nearly unlimited source material from samples of interest. LCLs are used to address questions in population genomics, mechanisms of disease, and pharmacogenomics. Thus, it is of fundamental importance to define the extent of chromosomal variation in LCLs. We measured variation in gen...

Characterization of Nucleoprotein Extracted from Human Influenza A Virus Cultured in Two Different Cell Lines

Background and Aims: Influenza virus nucleoprotein (NP) has the capacity to be used as subunit vaccine, but little is known about the impact of different cultures on its structure. In the present study we aimed to evaluate and compare the Isoelectric focusing (IEF) property of extracted viral nucleoproteins derived from Madin Darby canine kidney (MDCK) cell line and embryonated chicken eggs (EC...

فراوانی آسیب‌های کروموزومی ناشی از داروی بلئومایسین سولفات در سلول‌های هیبریدومای F3B6 و HF2x653 در مقایسه با سلول‌های غیرهیبرید والدی

Background and Objective: Hybrid cells are made from fusion of two or more somatic cells. After formation chromosomes are located in one membrane . . So nucleic condition of each fused cell has changes and two genomes and chromosomes interact with each other. Locating the genes in new nucleic and cytoplasmic condition and great chromosomal rearrangement in these new formed cells can affect th...

comparison of two different immature mouse ovarian vitrification methods using trypan blue staining

objective: different cryoprotectants are used for cryopreservation of ovarian tissue in patients at risk of infertility. ethylene glycol (eg), dimethyl sulfoxide (dmso) and propanediol (proh) have been chosen as the basic permeable cryoprotectants due to their decreased glass-formation characteristics compared to other cryoprotectants. in the present study, the effects of two different vitrific...
